Spray drying is a precedented method for the continuous manufacturing of engineered dry powder particles from a liquid feed solution. The process is readily tunable and scalable which allows for rapid formulation screening, using a small amount of active, while also directly translatable to cGMP manufacture. Given these attributes the technology is applicable to formulating and processing biotherapeutic products.

A large fraction of new chemical entities require solubilized formulations to achieve efficacious oral exposure. Amorphous solid dispersions, typically made by hot melt extrusion or spray drying, increase dissolution rate and supersaturated concentrations of these insoluble compounds in the gastrointestinal tract, which often translates to rapid absorption and improved bioavailability.

This webinar will focus on:

  • Spray-dried dispersion-based formulations of low-solubility compounds, including an overview of the formulation process, and rapid performance screening tests for guiding selection of compositions.
  • Multicomponent amorphous formulations, in which two or more excipients are formulated with the API, can be valuable in formulating challenging compounds.
  • The design of multicomponent amorphous compositions and architectures for simultaneously achieving manufacturability, physical stability, rapid dissolution and sustainment of supersaturated drug concentrations.
